68Ga-NODAGA-ZIGF-1R:4:40 is a radiolabeled PET imaging agent designed for the non-invasive detection of Insulin-like Growth Factor 1 Receptor (IGF-1R) expression in malignant tumors. The molecule consists of an engineered protein scaffold (Affibody), designated ZIGF-1R:4:40, which is derived from the B-domain of staphylococcal protein A and selected for its high affinity and specificity to the extracellular domain of IGF-1R. This affibody is site-specifically conjugated to the chelator NODAGA (1,4,7-triazacyclononane,1-glutaric acid-4,7-acetic acid) and labeled with the positron-emitting radioisotope Gallium-68 (68Ga). IGF-1R is a transmembrane tyrosine kinase overexpressed in several cancers, including glioma, lung, and colorectal cancer, where it drives proliferation and therapy resistance. By binding to the receptor with sub-nanomolar affinity, this tracer enables patient stratification for IGF-1R-targeted therapies and facilitates real-time monitoring of receptor expression status throughout the course of treatment.
