Overview

7-difluoromethoxyl-5,4'-di-n-octylgenistein (DFOG) is a novel synthetic analogue of genistein designed to enhance anti-cancer activity. It is a small molecule isoflavone derivative with difluoromethoxy and di-n-octyl substitutions that increase its potency compared to natural genistein. DFOG has demonstrated significant inhibitory effects on the proliferation and self-renewal capacity of cancer stem-like cells in ovarian cancer by downregulating the expression of forkhead box M1 (FOXM1), a transcription factor associated with tumorigenicity and stemness. It also inhibits growth in gastric cancer cell lines through similar mechanisms involving cell cycle arrest and modulation of key regulatory proteins such as CDK1, Cdc25B, cyclin B, and p27 KIP1[1][2][3]. The compound was developed for research purposes by academic groups in China.
