89Zr-Lumi804-Anti-PSMA + 227Th-Lumi804-Anti-PSMA is a preclinical radiotheranostic pair developed by Lumiphore for the imaging and treatment of prostate cancer. The system utilizes an anti-prostate-specific membrane antigen (PSMA) monoclonal antibody conjugated to Lumiphore's proprietary Lumi804 macrocyclic bifunctional chelator. The Lumi804 chelator is an octadentate ligand based on 1-hydroxypyridin-2-one (HOPO) units, designed to stably bind various radiometals at room temperature. In this matched pair, the antibody is labeled with Zirconium-89 (89Zr) for positron emission tomography (PET) imaging to identify and localize PSMA-expressing tumors, and with Thorium-227 (227Th) for targeted alpha therapy (TAT) to deliver high-energy alpha radiation directly to the cancer cells. This approach allows for personalized medicine by using the same targeting vector for both diagnostic imaging and therapeutic intervention.
