9-ING-41 + carboplatin is an investigational combination therapy consisting of 9-ING-41, a potent and selective small molecule inhibitor of glycogen synthase kinase-3 beta (GSK-3β), and carboplatin, a platinum-based chemotherapy. 9-ING-41 has demonstrated antitumor activity via downregulation of the NF-κB pathway and suppression of NF-κB target genes such as cyclin D1, Bcl-2, XIAP, and Bcl-XL, leading to inhibition of tumor growth and enhanced apoptosis in models of solid tumors and lymphomas. It also has potential immune modulatory effects by reducing checkpoint protein expression and enhancing T-cell/NK-cell activity. Carboplatin disrupts DNA replication and induces apoptosis in dividing cells. This combination is being evaluated primarily in patients with advanced, recurrent, or metastatic salivary gland carcinomas, including adenoid cystic carcinoma, with IV administration.
