Overview

99mTc-HYNIC-D(TPPE) is a radiopharmaceutical imaging agent consisting of a retro-inverso peptidomimetic derivative (D(TPPE)) conjugated with hydrazinonicotinic acid (HYNIC), radiolabeled with technetium-99m (99mTc) via tricine/EDDA co-ligands for single-photon emission computed tomography (SPECT) tumor imaging. The D(TPPE) peptide is structurally engineered by replacing L-amino acids with D-amino acids and reversing the primary amino acid sequence of the parent EPPT peptide, which targets the tumor-associated antigen MUC1. The radiotracer demonstrates high affinity for MUC1, favorable metabolic stability, and improved tumor-to-nontarget ratios compared to the original EPPT peptide, allowing clearer visualization of HT29 colon cancer tumors in preclinical models. The agent is designed to specifically bind and image MUC1-overexpressing cancers, with primary research in colon cancer[1].
