99mTc-ZHER2:41071 is a second-generation affibody molecule labeled with the radionuclide technetium-99m, designed for molecular imaging of human epidermal growth factor receptor 2 (HER2) expression in tumors using single photon emission computed tomography (SPECT). The ZHER2:41071 scaffold has been engineered to improve stability, hydrophilicity, and storage properties compared to earlier generations. It binds specifically and with high affinity to HER2-expressing cells (affinity ~58 pM), enabling non-invasive visualization and quantification of HER2 status in cancers such as breast cancer. Preclinical and phase I clinical studies have shown that 99mTc-ZHER2:41071 provides excellent tumor-to-blood ratios, low renal uptake compared to previous probes, favorable dosimetry, and safety in humans. Its primary use is as a diagnostic radiopharmaceutical for stratifying patients who may benefit from HER2-targeted therapies[1][4][6].
