AAHI-SC2 + AAHI-SC3 is a combination vaccine candidate consisting of two self-amplifying RNA (saRNA) components developed by the Access to Advanced Health Institute (AAHI) in collaboration with ImmunityBio. The vaccine is designed as a heterologous boost for individuals previously vaccinated against or infected with COVID-19. The saRNA platform utilizes a viral replicase (typically from alphaviruses) to enable the RNA to replicate itself within host cells, allowing for significantly lower doses compared to conventional mRNA vaccines while maintaining high levels of antigen expression. AAHI-SC2 encodes the SARS-CoV-2 spike protein, while AAHI-SC3 is designed to broaden the immune response, typically by targeting the conserved nucleocapsid (N) protein to elicit robust T-cell mediated immunity. The combination is formulated with AAHI's proprietary Lipid Inorganic Nanoparticle (LION) delivery system, which acts as both a carrier and an adjuvant to enhance the immune response.
