Overview

This drug is a **combination therapy** consisting of AB-729 (Imdusiran), a GalNAc-conjugated small interfering ribonucleic acid (siRNA) therapeutic, and a nucleos(t)ide analogue (NA), typically used for the treatment of **chronic hepatitis B (CHB)**. AB-729 targets HBV-infected hepatocytes via N-acetylgalactosamine (GalNAc) conjugation and inhibits hepatitis B virus protein and antigen production including hepatitis B surface antigen (HBsAg), which may break immune tolerance and restore anti-HBV T-cell activity. The nucleos(t)ide analogue (e.g., tenofovir or entecavir) inhibits HBV replication by blocking the reverse transcriptase activity of viral polymerase. Together, this combination aims for profound and sustained decreases in HBsAg in CHB patients, with the goal of enabling functional cure through viral suppression and immune reawakening[1][5][6][7]. Clinical data indicate this regimen is well tolerated and leads to meaningful HBsAg and HBV DNA reductions[1][3][5][7].
