AB-836 + nucleos(t)ide analogue is a combination therapy containing AB-836, a potent and highly selective hepatitis B virus (HBV) capsid assembly modulator (CAM), together with a nucleos(t)ide analogue, typically standard-of-care agents like entecavir or tenofovir. AB-836 binds to HBV core protein, inhibiting pregenomic RNA (pgRNA) encapsidation and viral replication, as well as interfering with covalently closed circular DNA (cccDNA) formation. The addition of a nucleos(t)ide analogue inhibits the viral polymerase, leading to additive antiviral efficacy. AB-836 demonstrated substantial reductions in HBV DNA levels in clinical studies but was discontinued due to safety findings. The combination represents a mechanistically complementary approach for disrupting HBV replication and cccDNA establishment.
