Overview

Abacavir + efavirenz + didanosine is a combination of three antiretroviral drugs used as part of highly active antiretroviral therapy (HAART) for the treatment of HIV-1 infection. Each component acts via a distinct mechanism to inhibit HIV replication: abacavir and didanosine are nucleoside reverse transcriptase inhibitors (NRTIs), while efavirenz is a non-nucleoside reverse transcriptase inhibitor (NNRTI). The combination is used in adults, often as a second-line therapy for those failing previous regimens. All three agents are taken orally and can lead to distinct as well as overlapping toxicities, most notably hypersensitivity and gastrointestinal symptoms (abacavir), central nervous system effects and rash (efavirenz), and pancreatitis or peripheral neuropathy (didanosine). Formulations containing all three as a single pill are not marketed, so this combination is prescribed as individual agents. All are approved pharmaceuticals but the combination itself has no unique brand name and may be less commonly used in current guidelines due to toxicity and drug-drug interaction concerns[1][7].
