Drug intelligence / Profile preview
**Abacavir + lamivudine + zidovudine** is an oral fixed-dose antiretroviral combination marketed as **Trizivir**. It contains three synthetic nucleoside analogue reverse transcriptase inhibitors: abacavir, lamivudine, and zidovudine. After intracellular phosphorylation to active nucleotide analogues, the components competitively inhibit HIV-1 reverse transcriptase and terminate viral DNA-chain elongation, suppressing HIV replication. The combination is indicated for treatment of HIV-1 infection, either alone in appropriate patients or with other antiretroviral agents. The fixed-dose tablet contains abacavir 300 mg, lamivudine 150 mg, and zidovudine 300 mg. Important risks include potentially fatal abacavir hypersensitivity, particularly in people positive for HLA-B*5701; zidovudine-associated anemia, neutropenia, and myopathy; lactic acidosis/hepatomegaly with steatosis; and hepatitis B exacerbation after stopping lamivudine-containing treatment.
