Overview

ABBV-453 + daratumumab + lenalidomide + dexamethasone is an investigational four-drug combination for the treatment of relapsed or refractory multiple myeloma. ABBV-453 is an orally available investigational small-molecule inhibitor being developed by AbbVie, while daratumumab is a monoclonal antibody targeting CD38, lenalidomide is an immunomodulatory agent, and dexamethasone is a synthetic corticosteroid. Together, they target malignant plasma cells via multiple pathways: ABBV-453 (mechanism details undisclosed), daratumumab (CD38 inhibition and immune activation), lenalidomide (modulation of immune function and inhibition of angiogenesis), and dexamethasone (anti-inflammatory and anti-myeloma effects). This combination is being studied for safety and preliminary efficacy in patients with multiple myeloma[3][7].
