ABBV-621 + FOLFIRI is a combination therapy consisting of the investigational biologic ABBV-621 (eftozanermin alfa), a TRAIL receptor agonist, and the established chemotherapy regimen FOLFIRI, which includes folinic acid (leucovorin), fluorouracil (5-FU), and irinotecan. ABBV-621 is designed to activate the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) pathway, inducing apoptosis in cancer cells. FOLFIRI disrupts cancer cell proliferation through DNA synthesis inhibition and topoisomerase I inhibition. This combination has demonstrated synergistic anti-tumor activity in preclinical models and is being studied especially for metastatic colorectal cancer and other solid tumors[1][3][5].
