abemaciclib + abiraterone

**abemaciclib + abiraterone** is an investigational oral combination therapy used in clinical trials for metastatic castration-resistant prostate cancer (mCRPC). Abemaciclib is a selective inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), which blocks cell cycle progression, while abiraterone inhibits the enzyme CYP17A1, leading to decreased androgen production and suppression of androgen receptor (AR) signaling, a central driver in prostate cancer. This combination is designed to target both the AR signaling pathway and cell cycle progression to overcome resistance mechanisms and improve outcomes in prostate cancer. The combination is studied with concomitant prednisone or prednisolone. The developer and funder of these studies is Eli Lilly and Company.
