Overview

This combination therapy involves the CDK4/6 inhibitor abemaciclib administered alongside androgen deprivation therapy (ADT) and radiation therapy. Abemaciclib, developed by Eli Lilly and Company, is an oral small molecule that targets cyclin-dependent kinases 4 and 6, which are critical regulators of the cell cycle progression from the G1 to S phase. In prostate cancer, androgen receptor signaling is known to activate the CDK4/6-Cyclin D complex to sustain cancer cell proliferation. The rationale for this combination is that inhibiting CDK4/6 may overcome resistance to hormonal therapy (ADT) and enhance the efficacy of radiation in patients with localized high-risk or locally advanced prostate cancer. The University of Alabama at Birmingham is conducting a Phase II trial to evaluate the safety and clinical response of this specific regimen.
