Overview

**Abemaciclib + iohexol** is a co-administration of two distinct drugs: **abemaciclib**, an oral small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6) developed for the treatment of hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative breast cancer, and **iohexol**, a non-ionic, water-soluble radiographic contrast agent used for measuring glomerular filtration rate (GFR) and in radiographic imaging. This combination is primarily used in clinical pharmacology studies to evaluate the effect of abemaciclib on renal function. Abemaciclib can increase serum creatinine by inhibiting renal transporters responsible for tubular secretion of creatinine, without affecting true GFR, as measured by iohexol clearance[1][3][4]. The investigational use of this combination is for assessment of renal transporter inhibition and validation of alternative renal function measurement in patients receiving abemaciclib[1][2][3]. Abemaciclib is developed and manufactured by Eli Lilly, and iohexol is developed and manufactured by GE Healthcare (Omnipaque).
