Abemaciclib + niraparib is an investigational oral combination therapy consisting of two small molecule drugs: abemaciclib, a selective cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor, and niraparib, a poly(ADP-ribose) polymerase (PARP) inhibitor. Abemaciclib inhibits CDK4/6 to block cell cycle progression in cancer cells, while niraparib impairs DNA repair by inhibiting PARP enzymes. The combination aims to enhance antitumor activity by simultaneously disrupting cell proliferation and DNA repair mechanisms. This regimen is being studied primarily for late-line treatment of ovarian cancer and as neoadjuvant therapy in hormone receptor-positive, HER2-negative breast cancer[1][2][3][4].
