Overview

Abemaciclib + pembrolizumab is an investigational combination therapy consisting of two distinct agents with complementary mechanisms of action. Abemaciclib is a selective small-molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), which blocks cell cycle progression in cancer cells by inhibiting phosphorylation of the retinoblastoma protein, thereby preventing cellular proliferation. Pembrolizumab is a humanized monoclonal antibody that targets the programmed death-1 (PD-1) receptor on T lymphocytes; it blocks PD-1 interaction with its ligands PD-L1 and PD-L2, reactivating T-cell–mediated immune responses against tumor cells. The combination has been studied in advanced cancers such as hormone receptor-positive/HER2-negative metastatic breast cancer, non-small cell lung cancer (NSCLC), and gastroesophageal adenocarcinoma to assess potential synergistic antitumor effects by combining targeted inhibition of cell cycle progression with immune checkpoint blockade[1][2][3][4][5].
