ABI-H0731 + nucleos(t)ide + peginterferon alfa-2a is a combination investigational therapy for chronic hepatitis B virus (HBV) infection. **ABI-H0731 (vebicorvir)** is a small-molecule direct-acting antiviral that inhibits the HBV core protein, interfering with pregenomic RNA encapsidation and DNA replication, and disrupts HBV nucleocapsids, reducing new cccDNA formation. **Nucleos(t)ide analogues** (nucleoside/nucleotide reverse transcriptase inhibitors; NrtIs) inhibit HBV polymerase/reverse transcriptase, suppressing viral DNA synthesis. **Peginterferon alfa-2a** is a pegylated recombinant interferon stimulating the innate antiviral immune response via the JAK/STAT pathway and interferon alpha/beta receptors. The combination is aimed at achieving deeper or more durable HBV suppression than monotherapy by targeting multiple HBV lifecycle steps and stimulating host immunity.
