ABI-H2158 + entecavir

**ABI-H2158 + entecavir** is an investigational oral combination therapy for chronic hepatitis B virus (HBV) infection. ABI-H2158 is a potent, direct-acting small-molecule HBV core protein inhibitor whose mechanism involves preventing HBV replication through dual effects: blocking pregenomic RNA (pgRNA) encapsidation and disassembling nucleocapsids, thus inhibiting both early and late HBV replication steps. Entecavir is a nucleoside analogue reverse transcriptase inhibitor (NrtI) approved for chronic HBV. Combined, these agents aim to achieve greater suppression of viral replication and antigen production compared to entecavir alone. Clinical development of the ABI-H2158 + entecavir regimen was terminated due to hepatotoxicity observed in the combination arm in Phase 2 studies[1][5][7].
