Drug intelligence / Profile preview
Abiraterone + dalpiciclib is an investigational combination regimen consisting of two small molecule drugs: abiraterone, a CYP17 inhibitor (androgen biosynthesis inhibitor), and dalpiciclib, a cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor. This dual-targeted approach is being evaluated in a prospective, open-label, single-arm Phase II clinical trial sponsored by Shanghai Ninth People's Hospital Affiliated to Shanghai Jiao Tong University for the treatment of androgen receptor (AR)-positive, HER2-negative recurrent or metastatic salivary gland carcinoma. By combining hormonal blockade (suppressing androgen synthesis and targeting AR signaling) with cell-cycle inhibition (blocking cell cycle progression), the regimen aims to synergistically inhibit tumor growth in patients whose tumors express the androgen receptor but lack HER2 amplification. Additionally, the combination is being investigated in Phase III trials for metastatic hormone-sensitive prostate cancer (mHSPC).
