Overview

ABL103 + pembrolizumab is a combination immunotherapy regimen under clinical investigation for advanced or metastatic solid tumors. ABL103 is a bispecific antibody developed by ABL Bio that targets B7-H4 and 4-1BB: it is designed to activate T cells selectively within the tumor microenvironment—leveraging the Grabody-T platform—to enhance antitumor responses while reducing off-tumor toxicity. Pembrolizumab (KEYTRUDA), developed by Merck & Co., Inc., is a monoclonal antibody that blocks the programmed cell death protein 1 (PD-1) receptor, unleashing T cell–mediated immune responses against tumors. The combination aims to synergistically increase antitumor immunity: ABL103 provides co-stimulatory T cell activation in B7-H4+ environments, while pembrolizumab relieves PD-1–mediated immune suppression. The combination is being evaluated in a Phase 1b/2 clinical trial for safety and efficacy in advanced/metastatic solid tumors, sometimes also in triple combination with taxane-based chemotherapy[1][2][3][4][5][6].
