Overview

ABP-671 + allopurinol is a combination therapy being developed by Atom Therapeutics for the treatment of chronic gout and hyperuricemia. ABP-671 (lingdolinurad) is a novel, potent, and selective oral small molecule inhibitor of the urate transporter 1 (URAT1), which works by reducing the renal reabsorption of uric acid. Allopurinol is a standard-of-care xanthine oxidase inhibitor that reduces the systemic production of uric acid. By combining these two distinct mechanisms—increasing renal excretion and decreasing production—the therapy aims to achieve more significant reductions in serum uric acid levels than monotherapy, particularly in patients who do not reach target levels on allopurinol alone. Clinical development includes drug-drug interaction studies to evaluate the safety and pharmacokinetic profile of ABP-671 when added to stable allopurinol therapy in gout patients, including those with renal impairment.
