ABS-101 (also known as TROP2 TCE T-MATE) is a preclinical-stage, conditionally activated T-cell engager (TCE) bispecific antibody developed by Amberstone Biosciences. Built on the proprietary T-MATE (Tumor Microenvironment Activated Therapeutic) platform, ABS-101 is designed as a 2+1 heterodimeric IgG1-based bispecific antibody that targets TROP2 (Trophoblast Cell Surface Antigen 2) on tumor cells and CD3 on T cells. The T-MATE platform utilizes a pH-dependent conformational change to selectively activate the TCE in the acidic tumor microenvironment (TME), thereby minimizing on-target, off-tumor toxicities in healthy tissues where TROP2 is normally expressed. In preclinical studies, ABS-101 demonstrated robust pH-selectivity, potent T-cell-dependent cytotoxicity, and significant anti-tumor activity in xenograft models of solid tumors, including ovarian, gastric, pancreatic, breast, and non-small cell lung cancers.
