ABSK061 + ABSK043 + CAPOX

ABSK061 + ABSK043 + CAPOX is an investigational combination therapy developed by Abbisko Therapeutics for the treatment of advanced solid tumors harboring FGFR2/3 alterations. The regimen consists of ABSK061, a highly selective, next-generation small molecule inhibitor of fibroblast growth factor receptors 2 and 3 (FGFR2/3); ABSK043, a first-in-class oral small molecule inhibitor of programmed death-ligand 1 (PD-L1); and the CAPOX chemotherapy regimen, which includes capecitabine and oxaliplatin. ABSK061 is designed to target tumors with FGFR2/3 mutations or fusions while minimizing activity against FGFR1 to reduce off-target side effects. ABSK043 works by binding to PD-L1 and inducing its internalization, effectively blocking the PD-1/PD-L1 immune checkpoint interaction. This combination is currently being evaluated in a Phase 2 clinical trial (NCT06632262) for patients with metastatic or unresectable solid tumors, including gastric, gastroesophageal junction, urothelial, and non-small cell lung cancers.
