ABT-348 is a potent, novel small molecule inhibitor that targets Aurora A, B, and C kinases as well as all members of the vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF) receptor tyrosine kinase families. It acts as an adenosine triphosphate competitive inhibitor and has demonstrated antitumor activity in preclinical cancer models. Docetaxel is a taxoid antineoplastic agent used in the treatment of various cancers including breast cancer, non-small cell lung cancer (NSCLC), prostate cancer, gastric adenocarcinoma, and head and neck squamous cell carcinoma. Docetaxel promotes microtubule assembly from tubulin dimers and stabilizes microtubules by preventing depolymerization, thereby inhibiting normal dynamic reorganization essential for mitosis. The combination of ABT-348 with docetaxel represents an investigational regimen aimed at enhancing anticancer efficacy through dual inhibition of mitotic kinases/microtubule stabilization.
