Overview

Acasunlimab is a bispecific monoclonal antibody (also known as GEN1046 or BNT311, and DuoBody-PD-L1x4-1BB) designed to elicit an antitumor immune response by conditionally activating 4-1BB only when bound to PD-L1. This mechanism aims to enhance T-cell mediated anti-tumor activity while minimizing off-target toxicity. Pembrolizumab (KEYTRUDA) is a monoclonal antibody that blocks the programmed cell death protein 1 (PD‑1), thereby preventing tumor cells from evading immune detection. The combination of acasunlimab and pembrolizumab, with or without chemotherapy, is being investigated for the treatment of metastatic non-small cell lung cancer (NSCLC), particularly in patients who have progressed after prior PD-(L)1 inhibitor and platinum-based chemotherapy[2][3][7]. Acasunlimab was developed by Genmab and BioNTech; pembrolizumab was developed by Merck & Co.
