Acetaminophen (also known as paracetamol) is a small molecule analgesic and antipyretic. This specific test formulation was developed by the McNeil Consumer Healthcare division of Johnson & Johnson to evaluate its efficacy and safety in treating acute postoperative dental pain. In a Phase 2 clinical trial (NCT02320708), a 1000 mg dose (administered as two 500 mg tablets) was compared against commercial acetaminophen, ibuprofen, and placebo following third molar extractions. While the precise mechanism of action of acetaminophen is not fully understood, it is primarily attributed to the inhibition of prostaglandin synthesis within the central nervous system, likely through the modulation of cyclooxygenase (COX) enzymes, and may also involve the activation of descending serotonergic pathways.
