Ivagan is a marketed fixed-dose combination drug developed by Biotech Laboratorios for the acute, symptomatic treatment of migraine attacks and severe vascular headaches. It contains three active ingredients: acetaminophen (paracetamol), caffeine, and dihydroergotamine mesilate. Acetaminophen acts as an analgesic and antipyretic by inhibiting prostaglandin synthesis in the central nervous system. Dihydroergotamine is an ergot alkaloid that acts as a selective agonist at 5-HT1B and 5-HT1D receptors, causing vasoconstriction of dilated intracranial extracerebral vessels and inhibiting trigeminal peptide release. Caffeine is a central nervous system stimulant and adenosine receptor antagonist that enhances the enteral absorption and analgesic efficacy of both acetaminophen and dihydroergotamine. Ivagan is approved and marketed in Venezuela as oral tablets.
