This is a combination drug consisting of three active pharmaceutical ingredients: - **Acetaminophen (paracetamol):** A non-opioid analgesic and antipyretic that works primarily by inhibiting prostaglandin synthesis in the central nervous system, leading to reduced pain and fever. - **Etoricoxib:** A selective cyclooxygenase-2 (COX-2) inhibitor classified as a nonsteroidal anti-inflammatory drug (NSAID). It reduces inflammation and pain by selectively blocking COX-2, an enzyme involved in prostaglandin production. - **Tramadol:** An opioid analgesic that acts as a weak agonist at the μ-opioid receptor and also inhibits reuptake of norepinephrine and serotonin, contributing to its analgesic effects. The rationale for combining these agents is to provide multimodal analgesia—targeting different pain pathways—to achieve superior pain relief with potentially lower doses of each component. This approach may reduce side effects associated with higher doses of individual drugs. While combinations such as acetaminophen/tramadol or etoricoxib/tramadol are studied for acute low back pain and other painful conditions[1][2][3][5], there is limited evidence or commercial availability for a fixed-dose triple combination containing all three agents together.
