Overview

This combination analgesic therapy consists of the central analgesic acetaminophen (paracetamol) and the nonsteroidal anti-inflammatory drug (NSAID) ketorolac tromethamine. It is being investigated by the Universidade Estadual de Ponta Grossa for the prevention of tooth sensitivity associated with in-office dental bleaching procedures using 35% hydrogen peroxide. Ketorolac tromethamine acts by inhibiting cyclooxygenase enzymes (COX-1 and COX-2), thereby reducing the synthesis of prostaglandins that mediate inflammation and pain, while acetaminophen provides complementary analgesic effects through central pathways. The treatment protocol involves oral administration starting one hour before the dental procedure and continuing every 8 hours for 48 hours.
