Overview

Acetaminophen + propofol is a combination of an analgesic/antipyretic and a sedative-hypnotic agent, frequently utilized in multimodal anesthesia protocols. Acetaminophen (paracetamol) exerts its effects primarily by inhibiting cyclooxygenase (COX) enzymes in the brain, thereby reducing prostaglandin synthesis, and may also interact with the endogenous cannabinoid and serotonergic systems. Propofol is a short-acting intravenous anesthetic that enhances the activity of gamma-aminobutyric acid (GABA) at the GABA-A receptor complex, leading to hyperpolarization of neurons and subsequent sedation or general anesthesia. The co-administration of these agents is intended to provide synergistic effects, often allowing for a reduction in the required dose of propofol (anesthetic-sparing effect) and improving the quality of postoperative recovery by providing early analgesia.
