Drug intelligence / Profile preview
Tramadol hydrochloride/Acetaminophen SR Tab. (marketed as Wontran ER) is a fixed-dose, sustained-release combination of tramadol hydrochloride, a centrally acting synthetic opioid analgesic, and acetaminophen, a non-opioid analgesic and antipyretic. Developed by Daewon Pharmaceutical, this formulation is designed to provide both rapid and prolonged pain relief by combining an immediate-release layer and a sustained-release layer of the active ingredients. Tramadol acts as a weak agonist at mu-opioid receptors and inhibits the reuptake of serotonin and norepinephrine, while acetaminophen is believed to act primarily in the central nervous system to inhibit prostaglandin synthesis. This synergistic combination is approved in South Korea for the management of moderate to severe acute and chronic pain, offering improved patient compliance and a reduced dosing frequency compared to immediate-release formulations.
