Overview

ACY-241 + paclitaxel is a combination regimen consisting of ACY-241 (citarinostat), a novel, orally available and selective histone deacetylase 6 (HDAC6) inhibitor, and paclitaxel, a standard chemotherapeutic agent that stabilizes microtubules. ACY-241 selectively inhibits HDAC6, leading to hyperacetylation of α-tubulin and altered microtubule dynamics, resulting in increased stability and apoptosis of cancer cells. Paclitaxel binds to tubulin and promotes microtubule assembly, preventing their depolymerization and leading to mitotic arrest and cell death. The combination synergistically enhances anti-tumor efficacy by increasing cancer cell apoptosis, promoting abnormal multipolar spindle formation, inducing cell cycle arrest, and increasing aneuploidy. This regimen has demonstrated significant tumor growth suppression and increased apoptotic markers in preclinical models of solid tumors (notably ovarian, pancreatic, and breast cancer) and is undergoing clinical development for patients with advanced solid tumors[1][2][3].
