ACY-241 + pomalidomide + dexamethasone is a combination therapy under clinical investigation, mainly for the treatment of relapsed or relapsed/refractory multiple myeloma. ACY-241 (also called citarinostat) is a second-generation, orally available, selective histone deacetylase 6 (HDAC6) inhibitor; pomalidomide is an immunomodulatory drug (IMiD); dexamethasone is a synthetic glucocorticoid corticosteroid. Together, this regimen seeks to leverage the synergy between HDAC6 inhibition and IMiD-induced cytotoxicity, promoting increased apoptosis and tumor growth inhibition with manageable toxicity profile. ACY-241 enhances anti-myeloma activity via epigenetic modulation, while pomalidomide modulates the immune microenvironment and dexamethasone adds anti-inflammatory and apoptotic effects. The combination has shown promising efficacy and safety in phase 1b studies in relapsed or refractory multiple myeloma[1][3][5].
