Overview

Ad[CE1A] is a research-stage oncolytic adenovirus engineered for the treatment of multiple myeloma. It achieves tumor selectivity by placing the expression of the essential viral replication gene E1A under the transcriptional control of the CS1 (also known as SLAMF7) promoter. This design ensures that the virus selectively replicates in and lyses cancer cells that express high levels of SLAMF7, a surface marker highly prevalent on multiple myeloma cells. Preclinical research, including a 2025 publication in the journal *Leukemia*, has demonstrated that Ad[CE1A] can effectively kill myeloma cells as a monotherapy and exhibits enhanced anti-tumor activity when combined with standard myeloma treatments such as proteasome inhibitors, alkylating agents, and histone deacetylase inhibitors.
