Overview

**Ad-p53 + capecitabine** is a combination of an investigational gene therapy (Ad-p53) and a cytotoxic chemotherapy agent (capecitabine). **Ad-p53** is a recombinant non-replicating adenovirus delivering a wild-type **TP53** gene to tumor cells via intratumoral injection, aiming to restore or supplement tumor suppressor p53 activity that is lost or mutated in many cancers. The transgene expresses functional p53 protein to promote cell cycle arrest, DNA repair, apoptosis, and immunogenic tumor cell death. **Capecitabine** is a prodrug of 5-fluorouracil (5-FU), an oral chemotherapy agent that inhibits thymidylate synthase, impairing DNA synthesis and leading to tumor cell death. This combination is primarily studied in advanced or refractory solid tumors to exploit potential synergy between restoration of p53-mediated cell death pathways and 5-FU-induced cytotoxicity.
