Ad-RTS-hIL-12 + veledimex is a novel gene therapy combination where Ad-RTS-hIL-12 is an inducible adenoviral vector engineered to express human interleukin-12 (IL-12) under the control of the RheoSwitch Therapeutic System® gene switch. Veledimex is an oral activator ligand that controls the expression of IL-12. When administered, veledimex crosses the blood-brain barrier and activates the gene switch, allowing for controlled production of IL-12, which enhances the immune system's ability to kill tumor cells by promoting activation of natural killer cells, inducing secretion of interferon-gamma, and potentially interfering with blood flow to tumors. This controlled immunogene therapy approach extends the IL-12 therapeutic window while managing toxicity.
