Ad26.Filo + MVA-BN-Filo is a heterologous prime-boost vaccine regimen developed by Janssen Vaccines in collaboration with Bavarian Nordic to provide broad protection against major filoviruses. The regimen consists of a priming dose, Ad26.Filo, which is a mixture of three recombinant adenovirus serotype 26 (Ad26) vectors, each expressing the glycoprotein (GP) of either Ebola virus (Zaire), Sudan virus, or Marburg virus. This is followed approximately 8 weeks later by a booster dose of MVA-BN-Filo (marketed as Mvabea), a recombinant, non-replicating Modified Vaccinia Ankara (MVA) vector. The MVA component encodes the glycoproteins of Ebola, Sudan, and Marburg viruses, as well as the nucleoprotein of the Taï Forest virus. This strategy is designed to elicit robust and durable humoral and cellular immune responses across multiple filovirus species. The regimen has been evaluated in Phase 1, 2, and 3 clinical trials and has demonstrated favorable safety and immunogenicity profiles in both adults and children.
