Ad26.ZEBOV + MVA-BN-Filo is a two-dose, heterologous Ebola vaccine regimen indicated for the prevention of disease caused by Zaire ebolavirus (Ebola virus) in individuals aged 1 year and older. The first dose, Ad26.ZEBOV (marketed as Zabdeno), is a monovalent, replication-incompetent adenovirus serotype 26 vector-based vaccine encoding the full-length glycoprotein of the Zaire ebolavirus Mayinga variant. The second dose, administered approximately eight weeks later, is MVA-BN-Filo (marketed as Mvabea), a recombinant non-replicating modified vaccinia Ankara vector-based multivalent filovirus vaccine encoding glycoproteins from Zaire ebolavirus (Mayinga variant), Sudan virus (Gulu variant), Marburg virus (Musoke variant), and nucleoprotein from Taï Forest virus. Both components are genetically modified so they cannot replicate in human cells but express viral proteins to stimulate an immune response. This regimen was authorized by the EMA in July 2020 and prequalified by WHO for use in outbreak settings and among at-risk populations including children and pregnant women[2][3][5][6][7].
