Overview

Ad35K++ is a synthetic recombinant protein derived from the adenovirus serotype 35 (Ad35) fiber knob, specifically engineered with mutations (D207G/T245A/I271V/F299Y) to significantly increase its binding affinity for CD46 (Membrane Cofactor Protein). Developed by Compliment, this protein acts as a CD46 antagonist. CD46 is a complement regulatory protein that is frequently overexpressed on various malignant cells, including those in B-cell non-Hodgkin's lymphoma, breast cancer, colon cancer, leukemia, and multiple myeloma. By overexpressing CD46, these tumors protect themselves from complement-dependent cytotoxicity (CDC) by inactivating complement components C3b and C4b. Ad35K++ binds to CD46 and blocks its ability to neutralize these complement factors, thereby sensitizing the tumor cells to complement-mediated attack. This mechanism is intended to enhance the efficacy of monoclonal antibody therapies or act as a standalone immunotherapy. The drug is currently in the preclinical stage of development.
