Ad5.F35-hGUCY2C-PADRE + Lm-GUCY2C is a heterologous prime-boost cancer vaccine regimen developed by Thomas Jefferson University for the treatment of advanced colorectal cancer and small bowel adenocarcinoma. The strategy utilizes two distinct platforms to deliver the tumor-associated antigen guanylyl cyclase C (GUCY2C), which is specifically expressed in the intestinal epithelium and overexpressed in gastrointestinal malignancies. The priming component, Ad5.F35-hGUCY2C-PADRE, is a chimeric adenoviral vector administered intramuscularly. The booster component, Lm-GUCY2C, is a recombinant, attenuated Listeria monocytogenes vector administered intravenously. This combination approach is designed to elicit a robust and durable CD8+ T-cell response against GUCY2C-positive tumor cells, overcoming the immune tolerance and waning efficacy often associated with single-platform viral vector vaccines.
