Overview

Ad6NSmut + AdCh3NSmut is a combination of two recombinant, replication-incompetent adenoviral vector vaccines designed for therapeutic immunization against hepatitis C virus (HCV). Both vectors are engineered to express a mutated nonstructural (NS) region of HCV, which is rich in conserved CD8+ and CD4+ T cell epitopes. The NS region includes a genetically inactivated NS5B polymerase. The combination leverages heterologous prime-boost vaccination to enhance the immune response and overcome pre-existing immunity to adenoviruses. These vaccines are derived from human (Ad6) and chimpanzee (AdCh3) adenovirus backbones, both modified for safety and immunogenicity. Clinical studies have shown that this regimen is well tolerated and capable of inducing robust HCV-specific T cell responses[1][2][5][6].
