Ad6NSmut + MVA-NSmut is an investigational therapeutic and prophylactic vaccine regimen for hepatitis C virus (HCV) infection. The approach uses a prime/boost strategy with two different viral vectors encoding nonstructural HCV proteins (genotype 1b, NS3–5B, mutated in NS5B to abolish polymerase activity). Ad6NSmut is a recombinant adenovirus serotype 6 vector, while MVA-NSmut is a modified vaccinia Ankara vector; both deliver the same HCV antigens. The regimen aims to induce robust and sustained T cell responses against HCV by first priming with the adenoviral vector and then boosting with the MVA vector. This strategy has been shown in clinical trials to generate strong CD4+ and CD8+ T cell responses targeting multiple HCV antigens, though efficacy in preventing chronic infection has not been demonstrated. The vaccines were initially developed by University of Oxford/Okairos AG and further developed by ReiThera and GSK[1][3][4][5][7].
