adagrasib + gemfibrozil

Adagrasib + gemfibrozil is a hypothetical or off-label combination of adagrasib, an oral small molecule KRAS G12C inhibitor indicated for adult patients with KRAS G12C-mutated locally advanced or metastatic non-small cell lung cancer (NSCLC) or colorectal cancer (typically with cetuximab), and gemfibrozil, an oral lipid-lowering agent used to treat hyperlipidemia by activating PPAR-alpha and reducing triglycerides. Adagrasib selectively and irreversibly inhibits the KRAS G12C mutant protein, blocking oncogenic signaling, whereas gemfibrozil acts primarily as a peroxisome proliferator-activated receptor alpha (PPARα) agonist, modifying lipid metabolism. No clinical evidence, regulatory approval, or dedicated studies exist for the combination of adagrasib and gemfibrozil, and there is no recognized indication for their combined use.
