adagrasib + nivolumab

Adagrasib + nivolumab is an investigational combination therapy consisting of adagrasib, a KRAS G12C inhibitor, and nivolumab, an anti-PD-1 immune checkpoint inhibitor. Adagrasib (Krazati) is a small molecule inhibitor that selectively targets KRAS G12C mutations, producing potent inhibition of KRAS-dependent signal transduction. Nivolumab (Opdivo) is an FDA-approved monoclonal antibody that blocks PD-1 receptors on T cells, enhancing anti-tumor immune responses. The combination is being studied primarily in non-small cell lung cancer (NSCLC) with KRAS G12C mutations, with the rationale that adagrasib may recondition the tumor immune microenvironment to become more immune-sensitive, potentially enhancing the efficacy of nivolumab.
