adagrasib + pembrolizumab

A combination therapy consisting of adagrasib (Krazati), a KRAS G12C inhibitor, and pembrolizumab (Keytruda), a PD-1 inhibitor. This combination is being investigated as a treatment for KRAS G12C-mutated non-small cell lung cancer (NSCLC), particularly in treatment-naïve patients. Clinical trials have shown promising efficacy and a manageable safety profile, with recent data presented at ASCO 2025 showing continued efficacy, especially in patients with PD-L1 TPS of 50% or higher. A phase 3 trial evaluating this combination versus pembrolizumab alone in treatment-naïve KRAS G12C-mutated NSCLC patients has been initiated.
