Overview

This is a combination of four tumor necrosis factor alpha (TNF-α) inhibitors—adalimumab, etanercept, golimumab, and infliximab. Each is a biologic agent used to treat various autoimmune and inflammatory diseases by inhibiting the activity of TNF-α, a key pro-inflammatory cytokine involved in the pathogenesis of conditions such as rheumatoid arthritis, psoriatic arthritis, ankylosing spondylitis, Crohn's disease, ulcerative colitis, and plaque psoriasis. These agents are typically used individually or in combination with other immunosuppressive drugs but are not standardly combined together due to overlapping mechanisms and increased risk of adverse effects. All four drugs act by binding to TNF-α and preventing it from activating its receptor on cell surfaces; however, their molecular structures differ (monoclonal antibodies vs. fusion protein). They have been developed by different pharmaceutical companies for similar indications[1][2][4][7][8].
