adapalene + doxycycline

A combination therapy consisting of topical adapalene 0.1% gel and oral doxycycline 100 mg/day, utilized as the conventional therapy comparator in clinical research conducted by the Pontificia Universidad Católica de Chile. Adapalene is a naphthoic acid derivative with retinoid activity that selectively binds to nuclear retinoic acid receptors (RAR-beta and RAR-gamma), leading to the normalization of follicular epithelial cell differentiation and reduction of comedone formation. Doxycycline is a broad-spectrum tetracycline antibiotic that inhibits protein synthesis by binding to the 30S ribosomal subunit of susceptible bacteria, including Cutibacterium acnes, and provides additional anti-inflammatory benefits by inhibiting neutrophil chemotaxis and matrix metalloproteinases. This combination is a standard-of-care approach for moderate inflammatory acne vulgaris.
