ADAPT-001 + checkpoint inhibitor is a combination therapy consisting of ADAPT-001, a replication-competent oncolytic adenovirus engineered to express a transforming growth factor beta (TGF-β) trap, and an immune checkpoint inhibitor (most commonly PD-1, PD-L1, or CTLA-4 monoclonal antibody). ADAPT-001 selectively infects and lyses tumor cells and locally secretes the TGF-β trap, which neutralizes the immunosuppressive impact of TGF-β within the tumor microenvironment. The checkpoint inhibitor component blocks immune checkpoints (such as PD-1/PD-L1), unleashing anti-tumor T cell responses. The combination is being developed to overcome resistance to immune checkpoint inhibitors in patients with advanced, refractory solid tumors including sarcoma, breast cancer, head and neck cancer, melanoma, and triple-negative breast cancer, with encouraging response rates and a favorable safety profile in early clinical trials.
